It is generally accepted that prion infection is due solely to a protein i.e. the protein-only hypothesis. The essential constituent of infectious prions is the scrapie prion protein (PrPSc) which is chemically indistinguishable from the normal, cellular protein (PrPC) but exhibits distinct secondary and tertiary structure. This very unusual feature seems to be in contradiction with a major paradigm of present structural biology stated by Anfinsen: a protein folds to the most stable conformation, this means only one structure.In order to (...) reconcile the results obtained on prions with the biophysics of protein folding, a model is proposed. It is based on the hypothesis that a thermodynamically irreversible step is involved in protein folding. The model is then extended to chaperone-assisted protein folding. It is shown that, under certain conditions, the transitory secondary structure formed during the earlier step of folding could interact with chaperone. Analysis shows that chaperone may help the protein to find correct conformation. On the other hand, analysis reveals the possibility that more than one structure may form from a single polypeptide chain. Under these conditions, the behaviour of chaperones resembles the characteristics of prion diseases. (shrink)

Context: Strict finitism is usually not taken seriously as a possible view on what mathematics is and how it functions. This is due mainly to unfamiliarity with the topic. Problem: First, it is necessary to present a “decent” history of strict finitism (which is now lacking) and, secondly, to show that common counterarguments against strict finitism can be properly addressed and refuted. Method: For the historical part, the historical material is situated in a broader context, and for the argumentative part, (...) an evaluation of arguments and counterarguments is presented. Results: The main result is that strict finitism is indeed a viable option, next to other constructive approaches, in (the foundations of) mathematics. Implications: Arguing for strict finitism is more complex than is usually thought. For future research, strict finitist mathematics itself needs to be written out in more detail to increase its credibility. In as far as strict finitism is a viable option, it will change our views on such “classics” as the platonist-constructivist discussion, the discovery-construction debate and the mysterious applicability problem (why is mathematics so successful in its applications?). Constructivist content: Strict finitism starts from the idea that counting is an act of labeling, hence the mathematician is an active subject right from the start. It differs from other constructivist views in that the finite limitations of the human subject are taken into account. (shrink)

Discusssion of passages 243 et. seq. of Wittgenstein's Philosophical lnvestigations tends to concentrate on the argument supporting the thesis that a logically private language is impossible. When the discussion becomes broader, the presumption is generally that this thesis is one premifs of an argument against solipsism. I believe that the passages will support a valid argument that might, at first glance, give comfort to someone in the egocentric predicament, but that this comfort would quickly grow cold on closer examination. I (...) do not mean to suggest that Wittgenstein offers no escape from the predicament, only that the escape will not be a consequence of the pluralism necessary to the existence of language, but rather a byproduct of his discussion of the logic of mental discourse. In this paper, I state an alternative to the contra-solipsist reading of the passages, taking Wittgenstein there to be reflecting a three part dispute, the other participants of which are a skeptic and a philosopher I'll call ‘the Realist'. The most recent commentator on this line, D. R. Shackleton Bailey, states that ‘spiritus is breath rather than odour’ and he has the support of some commentators, Marcilius, for example, who amends notus to motus, and Hertzberg, who takes it as sweet breath, citing Mart. 3. 65. 1. So also most translators : an exception is D. Paganelli who translates ‘aucun souffle, aucune odeur d'adultère’. However, the parallels cited by Shackleton Bailey are irrelevant to this situation: Afranius 243, Ach. Tat. (...) 2. 37. 9, and Claud. Carm. Min. 29. 33 all refer to the period just before, during, or immediately after the sexual act. It is most unlikely that this is the case in Propertius' poem. Propertius has come to see if Cynthia has spent the night alone; it is not a question of catching her inflagrante delicto, but of finding some rival there or the evidence of his stay not yet removed. Cynthia is indignant at her lover's suspicions and lest he should think that his rival had left earlier, she coarsely specifies the evidence that Propertius might expect to find, including spiritus admisso notus adulterio. The Dutch Reformed Church had a strong missionary DNA since the planting of the church. This missionary focus and fervour, however, ebbed and flowed during the history of the church. Saayman described that the mission upsurges in the DRC in four waves or 'periods of extraordinary mission endeavour'. The current research aimed to develop this theory through a literature study on the sociopolitical context and the developments in the ecclesiology and missiology of the DRC since the planting of the church (...) up to 2013. The research found strong evidence to define a fifth wave. The fifth wave was influenced by contextual changes and loss of influence and money by the DRC. Furthermore, the growth in the church's missional identity can be seen in the following aspects: the influence of theologians like Lesslie Newbigin and David Bosch; a belief that mission is not something the church does but something the church is; a shift from a Christocentric theology to a Trinitarian theology; a holistic view of salvation; a commitment to the local context of the congregation and a focus on bringing healing to their local communities; and lastly, the success of the Gestuurde Gemeentes network and, more recently, the Mission Shaped Ministry training.
